As we age, our skeletal muscle function and metabolism decline. The molecular processes underlying and the role physical activity plays in age-related decline are largely unknown. However, in a groundbreaking study, researchers examined muscle metabolism in both young and older adults, including those who exercised regularly, had normal activity levels, or were physically disabled.

What they discovered was remarkable – the molecule nicotinamide adenine dinucleotide (NAD+), which is believed to play a critical role in aging, was significantly lower in older adults, especially those with impaired physical function. A positive finding was that older adults who were active and exercised had NAD+ levels similar to those of younger individuals. It was also found that higher levels of NAD+ correlated with better mitochondrial and muscle function. These results suggest a clear connection between NAD+ and healthy aging in humans.
